中文摘要: 天然产物RP-66453属于一类具有良好生物活性的大环多肽类分子,它由二苯醚结构连接的肽环和联二苯酚结构连接的肽环构成,而且,由于RP-66453分子在受热时会转变成为热力学上更加稳定的阻转异构体,所以它的合成具有很大的挑战性。本项目拟采用仿生合成的设计思路,从简单化合物L-tyrosine和(S,S)-isoleucine出发,利用L-tyrosine中的苯酚基团,通过苯酚氧化偶联反应依次构建二苯醚桥联的肽环和联二苯酚桥联的肽环来合成RP-66453的双环多肽骨架,同时研究反应过程中的阻转异构选择性,最终实现RP-66453分子的合成。对于关键的苯酚氧化偶联反应,本项目拟通过中心金属Fe、Cu、Mn等与多齿氮配体或氧配体的相互作用搭建模拟细胞生物氧化酶的催化体系,通过清洁的氧化剂(氧气或双氧水等)来实现苯酚之间C-O键或C-C键的连接。

英文摘要: Natural product RP-66453 containing biaryl ether linkage and biphenol linkage belongs to a kind of macrocyclic peptides which have excellent bioactivities. When RP-66453 is heated, it transfers to its atropdiastereomer which is more thermodynamically stable, so it's quite challenging to synthesize RP-66453. This project will take a biomimetic synthetic strategy, starting from very simple molecules (L-tyrosine and (S,S)-isoleucine), taking advantages of the phenol group in L-tyrosine, building up peptide cycles containing biaryl ether linkage and biphenol linkage in succession to form the bicyclic peptides skeleton of RP-66453, researching the atropdiastereoselectivity in the reactions to realize the synthesis of RP-66453 finally. For the very important oxidative phenol coupling steps, this project will construct the catalytic systems by using metals (Fe, Cu, Mn etc.) and multidentate ligands containing nitrogen or oxygen atoms to simulate oxygenases in cell, and realize the linkages of phenol parts with C-O bond or C-C bond by using the clean oxidants (Oxygen gas, Hydrogen peroxide etc.).
