中文摘要: 单细胞凝胶电泳技术是一种快速、灵敏的检测细胞DNA 损伤与修复的方法,这一技术应用于环境生物监测时具有优于微核实验等技术的特点。将改良的SCGE技术与本地区海洋常见生物类群结合起来,找出符合本地地域特色的海洋环境污染SCGE生态毒理学监测途径;筛选合适的海洋生活单胞藻做为SCGE实验材料用于基因毒性、生态学、遗传毒理学、DNA 损伤与修复的检测等多方面研究。选择本地域常见多种无脊椎动物及鱼类,用于SCGE实验,检测环境污染物对动物的影响,以及这种影响与动物种类、性别、食性、环境温度以及采集地点之间的关系。为海洋环境生物监测进行综合分析提供科学的实验依据。

英文摘要: The single cell gel electrophoresis technique (SCGE or comet assay) is a simple, sensitive, reliable, and rapid method to detect DNA damage in eukaryotic cells. SCGE has advantages over other DNA damage methods, such as micronucleus assay. The improved SCGE was applied to the ecotoxicology monitoring of the local marine species. Suitable unicellular algae were used as SCGE experiment materials for genotoxicity, ecology, genetic toxicology, DNA damage and repair of testing. In addition, in various marine invertebrates and fish was used for the SCGE, to evaluate genotoxicity of pollutant and its differences between species, sex, diet, marine areas temperature, and sampled marine sites. The study provides date useful for integrated assessment of marine biological monitoring.
