中文摘要: 颅内动脉瘤破裂会引起严重的神经系统损害和不良预后。其病后复杂的病理生理机制仍然不清楚。我们的前期工作通过SOLEXA测序方法检出破裂动脉瘤患者血浆中15种异常表达谱的miRNA,提示血浆中miRNA表达谱改变可能作为此类疾病的生物标记物。我们将利用破裂颅内动脉瘤患者与非动脉瘤性脑出血及正常健康组相比较,增大样本例数(1000例)通过RCT-PCR验证并筛选2-5种动脉瘤破裂后血浆中特异表达的miRNA,明确其敏感及特异性;同时检测筛选出的候选miRNA是否在手术中获得的血管组织中有异常表达;在动脉瘤细胞株模型上,利用转染或阻断技术改变下调或上调miRNA的表达,寻求其下游可能调节的mRNA或蛋白的改变。目的是明确特异表达的miRNA 作为颅内动脉瘤破裂后的循环生物标记物或表达谱的作用,为研究颅内动脉瘤发生的病因、机制及可能的治疗靶点奠定理论基础。

英文摘要: intracranial aneurysm rupture is severe neurologic disorder and cause poor outcome.It is still unclear of its mechanism. we observes significant plasma changes of 15 miRNA based on SOLEXA process, indicate that these might be the potential biomarkers for disgonosing anenrysm.the aim is to compare the aneurysm reptured patients to those who are intracranial hemorrhage whithout aneurysm and healthy placebo, increase the sample to 1000 cases. try to select 2-5 miRNA via RCT-PCR which is significant different. to evaluate its specility and sensitivity. Figure out the mRNA or proteins on which might be worked. to find out the target point of either the possible mechanism and treatment.
