中文摘要: 综合运用并借鉴生化分析领域各学科新技术及研究方法,以多种环境友好的介质为原料,结合仿生分子识别、靶向激活荧光成像探针、细胞生理状态分子探针等技术,合成系列生物相容性良好的纳米胶囊。在此基础上构建具有癌细胞荧光成像检测、抗癌药物靶向投送、细胞生理状态指示等功能的纳米胶囊探针。利用激光共聚焦、全内反射技术等检测手段研究纳米胶囊靶向给药、细胞生理动态荧光标记等与癌细胞的生理相关的动态作用过程,并以之为基础建立灵敏的癌细胞检测新方法和有效的药物靶向投送新技术。为癌症的早期诊断、靶向药物的研制提供一定的理论基础。

英文摘要: Biosensors are developed for the study of the structure, the function and the molecular interaction of molecules in organisms, which plays important roles in the development of clinical, pharmaceutical, and environmental applications. Aptamer recognition with multifunctional nanocapsules with excellent properties, several novel bio-functionalized aptamer/nanocapsules probes were constructed for the specific recognition and detection of cancer cell. The experimental protocols could be summarized as follows: Acted as a cage-type cellular probe, several supramolecular aptamer/nanocapsules probes will develop for the high-intensity fluorescence microscopy imaging of cancer cells. The supramolecular aptamer/nanocapsules are construct from layer-by-layer self-assembly of aptamer/QD probes and nanocapsules functionalized with cancer cell-binding aptamer. Given new sensitive methods of detection of cancer cells. This strategy presents a promising platform for convenient evaluation of cancer cell.
