中文摘要: 骨髓间充质干细胞(MSCs)是一种具有多分化潜能的成体干细胞。体外试验证实MSCs在特定条件下可分化成为肺部上皮细胞,但关于MSCs在肺部生理及病理状态下气道上皮及肺实质的修复中所起作用的研究几近空白。本研究拟以绿色荧光蛋白C57BL/6转基因小鼠MSCs为供体细胞,以弹性蛋白酶肺泡灌洗致慢性阻塞性肺病(COPD)模型的同系小鼠为受体进行MSCs移植(以尾静脉注射和局部肺泡灌洗两种方式),用免疫组化方法和免疫透视电镜观察移植后MSCs在受体鼠肺脏各级结构的局部分化;评价细胞移植后对COPD模型鼠病理症状的改善情况;并对尾静脉注射和局部肺泡灌洗两种方式移植效率和综合效益进行比较分析。本研究的成功开展将有助于明确在肺部病理状态下细胞组织修复中的作用,为应用MSCs移植治疗广泛的肺部损伤性疾病提供实验依据。

英文摘要: mesenchymal stem cells (MSCs) as a kind of adult stem cell have the potency of multi-directional differentiation. In vitro, MSCs can differentiate into lung epithelial cells in certain condition. However, few studies were involved in physiological and pathological repair of airway epithelium and lung parenchymal cells by MSCs. In our study, GFP-MSCs isolated from C57BC/6 will be transplanted into COPD model lavaged with elastase by tail venous and .locally alveolar injection. Immunohistochemistry and electron microscope will be used to observe differentiation by MSCs in lung tissue, and improvement in pathology and presentation will be analyzed after transplantation and the engraftment efficiency and comprehensive effect will be compared between venous and intratracheal transplantation .This research will helpful explicit repair effect of MSCs in pathological lung condition and might provide experimental experience in future use.
