中文摘要: DNA甲基化和密切相关的储藏蛋白特异转录因子TaSPA、TaPBF和TaGAMYB参与谷蛋白家族基因表达的调控,但它们能否协同作用实现对该家族不同成员的多样性调控还不清楚。本课题拟检测小麦非种子器官/不同发育时期籽粒中高分子量麦谷蛋白基因甲基化的动态变化特征,通过与其表达水平的相关性分析,探讨甲基化对小麦高分子量麦谷蛋白编码基因表达的影响;筛选可能影响高分子量谷蛋白亚基基因甲基化修饰的甲基化/去甲基化酶,分析它们对麦谷蛋白基因甲基化修饰的调控特征;比较TaSPA、TaPBF和TaGAMYB与不同甲基化修饰水平的高分子量谷蛋白基因结合能力的差异,初步探索甲基化及TaSPA、TaPBF和TaGAMYB在谷蛋白基因表达中是否协同调控作用及其可能的机制。本课题预期结果将为进一步探讨小麦谷蛋白基因表达抑制/激活机制提供新的认识,并可为小麦品质改良提供依据。

英文摘要: DNA methylation and transcriptional factors TaSPA/TaPBF/TaGAMYB are claimed to participate in regulating the gene expression of wheat high molecular weight gluten subunit (HMW-GS) sub-family, but the issue as to whether or not they could orchestrate the family is not well shaped. In this project, we propose to (1) study the difference in the methylation levels of HMW-GS genes in wheat non-seed organ and different developmental stages of grain as well as their transcription profiles to uncover the effect of DNA methylation on HMW-GS gene expression; (2) isolate the methytransferases/demethytransferases that influence DNA methylation level of HMW-GS gene, analyze their regulatory roles in HWM-GS gene expression; (3) compare the binding capacity of TaSPA/TaPBF/TaGAMYB to their targeting sequence with differential methylation levels, so as to realize whether or not the synergetic exerting porfermance of DNA methylation and TaSPA/TaPBF/TaGAMYB on HWM-GS gene expression, as well as the possible machanism. The perspective outcomes of this project will help us to get a further insight into the mechinaries for both supression and activiation of HWM-GS genes, and will also provide a new way for the quality improvement of wheat grains.
