中文摘要: 前期研究发现龙眼果肉粗蛋白多糖能调节机体免疫功能,且其4个主要级分的结构特征和体外免疫刺激活性呈显著差异,但级分体内吸收利用及免疫调节的构效机制尚不清楚。本项目拟建立龙眼蛋白多糖各级分的荧光标记及高效凝胶渗透色谱分析法,通过分析各级分灌胃后在小鼠血液、组织器官和排泄物中的浓度动力学参数及分子量分布,探明其体内吸收、分布、降解和排泄行为;通过分析各级分灌胃后对小鼠免疫器官指数、免疫细胞功能和血清细胞因子表达的影响,探讨其结构特征-吸收利用-免疫调节的体内构效关系,并明确其主活性级分;通过主活性级分的体外模拟孵育降解实验,分离制备各降解级分,表征其一级和高级结构特征,分析其免疫细胞刺激活性,探讨结构变化对其体外免疫刺激的影响。研究揭示龙眼蛋白多糖结构与吸收利用和免疫调节的体内外构效机制,旨为龙眼精深加工提供理论依据,为果蔬多糖生物活性机制研究提供新思路。

英文摘要: The previous studies have confirmed that crude longan pulp proteoglycans possessed beneficial functions for mouse immune system, and their four purified factions displayed significant differences in structural feature and immunostimulatory activity in vitro. However, the uptake, utilization, immunomodulatory activity in vivo and potential structure-effect relationship of the polysaccharide factions remain unclear. The present research purposes to establish the microanalysis of longan proteoglycan fractions as fluorescent labeled derivatives by high performance gel permeation chromatography, measure the concentration related kinetic parameters and molecular weight distribution of the factions in mouse blood, tissue, organ and excretion after intragastric administration, and then reveal their in vivo behaviors including uptake, distribution, degradation and excretion. Moreover, the effects of peroral administrated longan proteoglycan fractions on the immune organ index, immunocyte function and cytokine secretion in serum of normal mice will be analyzed. Further, the in vivo relationships among the structure, uptake, utilization, and immunomodulatory activity for longan proteoglycan will be deduced, and the main active faction can be identified.
