中文摘要: 骨髓间充质干细胞龛(BMSCs niche)是以BMSCs为核心,以旁分泌和自分泌为主要调节手段的细胞局部"微环境"。既往研究表明补肾中药治疗骨质疏松的疗效发挥与调节BMSCs有关,但是国内外还没有从BMSCs龛的角度研究中药的作用机制。 本课题拟以补肾药治疗骨质疏松为切入点,在经典模型去卵巢大鼠模型上,通过micro-CT和骨组织形态计量学评价疗效,运用蛋白芯片、免疫组化、ELASA、Western blot、实时定量荧光PCR等技术,观察二仙汤对BMSCs龛的影响,聚焦共同参与调节龛环境的生物活性分子,观察其对BMSCs增殖、成骨分化功能的作用和作用机制,从而为中医"肾藏精,精生髓,髓生骨"理论提供部分科学阐释。

英文摘要: Bone mesenchymal stem cells niche (BMSCs niche)is a local "micro-environment" around BMSCs regulated by paracrine secretion and autocrine. Previous studies revealed the effect of traditional Chinese medicine effective in "invigorating the kidney" on osteoporosis is related to regulating BMSCs. However, little has been reported on the mechanism of TCM from the view of BMSCs niche. Present study is plan to observe the effect of Erxian Tang on classical osteoporosis rats model induced by OVX by evaluating bone mass density using micro-CT, exploring the bioactive factors in promoting BMSCs proliferation and differentiation using protein array, immunohistochemisty, ELISA, Western blot and real-time RT PCR techniques. We hope our study would contribute to the theory of "kidney storing essence, essence generating marrow, marrow producing bone" in TCM.
