中文摘要: 本课题旨在直接利用生物分子与稀土离子配位制备生物相容性好、发光效率高的空心多孔笼状稀土纳米配位聚合物(lanthanide nanoscale coordination polymers, Ln-NCPs),无需客体发光配体参与,阐明Ln-NCPs的形成机理及大小、厚度等参数与其荧光性能和分析灵敏度之间的关系,实现可控设计合成具有特定功能的Ln-NCPs,并以其为荧光探针通过时间分辨分析技术对细胞内的超氧自由基进行靶向监测,可望克服以往有机荧光探针生物相容性不好、水溶性差、易受背景荧光干扰等不足。超氧自由基靶向监测可用于揭示生物氧化衰老机理,为与活性氧相关疾病的诊断和治疗提供理论依据。本项目创新性地采用了空心多孔的笼状Ln-NCPs,充分利用了其比表面积利用率和荧光传感效率高的优势,进一步提高了其选择性和灵敏度,为稀土配位聚合物荧光探针在生物医学领域的应用提供了新思路。

英文摘要: This project is designed to prepare a series of hollow cage fluorescent lanthanide nanoscale coordination polymers (Ln-NCPs)by directly using biological molecules as bridge ligands, without auxiliary ligands to participate in to sensitize the fluorescence of lanthanide ions. Meanwhile, we will clarify the formation mechanism of Ln-NCPs,and demonstrate the relationship between parameters such as size, thickness with their fluorescence properties and detection sensitivity. These fluorescent Ln-NCPs exhibited excellent biocompatibility and highly emission efficiency were further used to tagert monitor the level of superoxide anion in cell. More important, the shortcomings of organic fluorescent probe, such as biocompatible and poor water soluble, tend to be interfered by background fluorescence, can be overcome in the mode of time-resolved fluorescence. Target monitoring the level of superoxide anion is benefit to reveal the biological oxidation mechanism and provide theoretical basis for the diagnosis and treatment of reactive oxygen species related diseases. Hollow cage Ln-NCPs with porous structure proposed here exhibit a higher accessible surface areas and excellent optical properity in sensing, comparing with compact pure ones. Therefore, they have a wider application in the biomedical field.
