中文摘要: 肿瘤良恶性的判断直接影响治疗方案的选择,目前影像学检查对部分病变的良恶性鉴别仍然存在困难。本项目拟构建hTERT启动子启动的铁蛋白基因载体,从体外(将该载体转染于良恶性肿瘤细胞)和活体(静脉注射该载体于良恶性肿瘤动物模型)两方面进行研究。采用MRI扫描于不同时间点观察体外细胞、活体肿瘤的信号变化,并对活体肿瘤信号变化区、体外细胞行RT-RCR、Western Blot、免疫组化、普鲁氏蓝染色以检测铁蛋白表达情况,对照MR信号改变特特点与组织病理检查结果。由于hTERT启动子是恶性肿瘤特异性启动子,只能在恶性肿瘤细胞内启动铁蛋白基因的表达并形成内源性的MRI对比剂,而在正常细胞及良性病变细胞处于关闭状态,不能形成对比剂,因此MRI一旦探测到信号变化即可反映肿瘤的恶性生物学行为。本项目研究思路完全不同于目前的靶向影像学研究思路,并有望为良恶性肿瘤的鉴别诊断建立一种新的广谱MRI诊断方法。

英文摘要: The evaluation of malignant behaviour of tumor determines the selection of treatment.Up to date, it is difficult to evaluate the malignant behaviour of tumor for imaging diagnosis. In the project,the Ferritin-gene vector mediated by hTERT promoter will be constructed. The vector will be transfected to benign and malignant tumor cells in vitro and be intravenously injected into rats with benign and malignant tumors in vivo. The tumor cells in vitro and tumors in vivo will be detected by MRI at different time point.The MRI signal changes will be analyzed. For the tumor cells in vitro and tumor tissue,the RT-PCR,WesternBlot,immunohistochemical staining and Prussian blue staining will be performed to test the expression of Ferritin gene.The ferritin expression will be compared with the MRI signal changes. hTERT promoter is the specific promoter of malignant tumor and will be mediated the expression of ferritin gene only in malignant tumor cells, not in normal cells and benign tumor cells,which will form the endogeneous MRI contrast agents.So, the MRI signal change will reflect the malignant behaviour of tumors. The research design is different from that of targeted imaging.
