中文摘要: 以在研青年基金研究工作为基础,本课题提出基于生物粘附作用构建一种具有叶酸-磁性双靶向、MR-NIR双模式成像、光动力-化学联合治疗的多功能上转换纳米复合材料。设想如下:通过多巴胺功能化的酞菁锌及多巴胺功能化的端基为叠氮的PEG-PCL在磁性上转换纳米粒子表面原位氧化聚合获得酞菁锌功能化的纳米粒子;通过Click反应将叶酸引入到粒子表面;利用粒子表面邻苯二酚与苯硼酸之间的共价作用实现对硼替佐米的负载。在肿瘤处放置磁铁,纳米粒子通过磁导靶向及叶酸介导靶在肿瘤处富集,在酸性的肿瘤中BTZ释放实现化学治疗;采用980 nm激光照射,650nm附近的发光激发酞菁锌分子产生单线态氧,实现光动力治疗,800nm附近的发光用于近红外成像,同时进行核磁共振成像。对该磁性上转换纳米材料在体内外的肿瘤诊疗方面的应用进行初步研究。期望通过本课题的研究能够为肿瘤诊疗用纳米材料的结构优化提供一些理论和实验依据。

英文摘要: According to the challenging problems in photodynamic therapy of tumors, based on the work for the youth fund research this project puts forward construction of a kind of multifunctional nanoparticles exhibit folic acid and magnetic target, MR@NIR dual modal imaging and PDT-chemotherapy functions based on dopamine bio-inspired adhesion interaction. The main assumptions are as follows: First the dopamine functionalized zinc phthalocyanine (ZnPc) and dopamine functionalized PEG-PCL with terminal azide group are in-situ polymerized on the surface of the magnetic upconversion nanoparticles. Then folic acid is grafted on the above nanoparticles through click reaction. Using the covalent-action between the catechol units and phenylboronic acid realize the loading of the chemotherapy drugs, bortezomib (BTZ). Put a magnet on the surface of the tumor, the multifunctional nanoparticles enrich in the tumor through magnetic targeting and folic acid mediated target, in the acidic environment of the tumor, BTZ release to achieve chemical treatment, at the same time by 980nm laser irradiation, 650nm near luminescence excite zinc phthalocyanine molecules to generate singlet oxygen, which realize the photodynamic therapy; in addition 800nm near the luminescence could be used for near infrared imaging, and magnetic resonance ima
