中文摘要: 重金属污染问题日趋严重,传统的重金属离子检测方法不能满足快速现场检测的需求。基于本课题组最近研制的能够在液相环境中工作的谐振式微悬臂梁高灵敏度质量型传感器,本研究首次尝试使用生物纳米材料-工程噬菌体作为特异性结合重金属离子的敏感层,发展一套对环境中痕量重金属离子快速、准确检测的新方法。利用金属硫蛋白特异性结合重金属离子的性质,通过分析比对蛋白质、核酸数据库中金属硫蛋白序列,初步筛选出可特异性结合重金属离子的保守5肽序列。应用M13噬菌体展示技术,在全部P8蛋白N末端融合表达这些5肽,构建工程噬菌体。这些工程噬菌体表面是一层金属硫蛋白5肽的单分子层,因此能够最大限度地特异性吸附重金属离子。应用谐振式微悬臂梁传感器筛选出与单一重金属离子特异结合的工程噬菌体,制造快速、灵敏、简便的可用于现场检测重金属离子的生物传感器。并丰富极具市场应用价值的金属硫蛋白与重金属离子结合活性的体外研究。

英文摘要: Heavy metal pollution has been an increasingly global problem for its danger to human health and to the environment. Traditional methods for detection of heavy metal ions are not suitable for on-spot application. Based on a newly develeped high quality-factor in-plane-mode resonant cantilever sensor for real-time bio/chemical detection in liquids, we will develop a nature nano-material (genetic engeered M13 bacteriophage) as the sensitive materials on the cantilever to detect the trace heavy metal ions in liquid. Metallothioneins, a family proteins with unclear biological function, have the capacity to bind heavy metal ions. Blast the amino acid sequence of the metallothioneins and select the conserved 5 peptides which possibly bind the heavy metal ions. Synthesis of DNA fragments with the sequence coding for 5 peptides in the P8 N-terminus and replace corresponding fragments into the genome of the M13 bacteriophage. The genetically engineered M13 bacteriophage would generate corresponding 5 peptides on their surface. Because of the largest heavy metal ion binding surface, the engineered M13 baceriophage could binds the ions as much as possible.
