中文摘要: 股骨头坏死塌陷后软骨损伤是亟待解决的临床难题,依靠骨髓基质干细胞(BMMSCs)构建的组织工程软骨为股骨头坏死塌陷后软骨损伤的修复带来了新机会。探索力学和基质微环境下BMMSCs向软骨细胞分化的调控及机制对于再生软骨质量控制具有重大意义。本研究以软骨细胞共培养诱导膜片化BMMSCs,结合含有多种内源生长因子的富血小板纤维蛋白(PRF),构建BMMSCs/PRF双膜复合体,模仿软骨生成的基质微环境。以此为基础,探讨不同压力作用下BMMSCs向软骨分化的生物力学特性,探索BMMSCs向软骨分化的力学信号转导机理。依靠将压力微环境调控的BMMSCs/PRF双膜复合体用于动物股骨头坏死塌陷后软骨的损伤修复,探索功能性力学作用与股骨头软骨生成间的量效关系。项目成果将为体内组织工程软骨的力学环境调控提供实验依据;为股骨头坏死塌陷后软骨损伤的治疗提供新思路和经验。

英文摘要: Cartilage injury postcollapse of osteonecrosis of the femoral head is an urgent clinical problem. We rely on bone marrow stromal cells ( BMMSCs ) to reconstruct tissue engineering cartilage,which brought new opportunity for repair of cartilage injury postcollpase of osteonecrosis of the femoral head. Exploring the mechanical and stromal microenvironment BMMSCs chondrogenic differentiation regulation and mechanism for regenerating cartilage quality control is of great significance. The study of cartilage cells induced by co-culture with diaphragm with BMMSCs, contains a variety of endogenous growth factor platelet rich fibrin ( PRF ). BMMSCs/PRF double membrane complex construction, simulation of cartilage regeneration microenvironment. On this basis, studies on the suitability of pressure under the stimulation of BMMSCs chondrogenic differentiation of biomechanical characteristics, exploring the BMMSCs chondrogenic differentiation of mechanical signal transduction mechanism. Relying on the pressure microenvironment preset BMMSCs/PRF double membrane complex for cartilage injury and repair postcollapse of osteonecrosis of the femoral head in animal.To explore functional mechanical stimulation and femoral head cartilage regeneration between the dose-effect relationship.
