中文摘要: 阿里红(Fomes officinalis Ames)是新疆维吾尔医常用药物,具有温肺化痰、活血消肿作用,其有效成分之一为多糖。本课题通过对维药阿里红中多糖提取分离和初步结构分析,测得阿里红粗多糖中还原糖含量为73.81 μ/ml, 糖醛酸含量为17.07%,单糖组成包括鼠李糖、阿拉伯糖、木糖、甘露糖、葡萄糖和半乳糖,其摩尔比为0.2:0.29:0.41:0.77:0.70:1.16。同时研究表明阿里红多糖是以未降解的药物的形式,主要是以被动扩散的方式被Caco-2细胞单层摄取和转运。在阿里红多糖干预小肠黏膜淋巴细胞因子表达的研究中,阿里红多糖可抑制小肠黏膜淋巴细胞数量,降低其分泌TNF-α12289;IL-1β27700;平,升高TGF-β27700;平,提示阿里红多糖可能通过抑制黏膜局部淋巴细胞的活性和炎症因子的表达,从而调节黏膜免疫状态。其它药效学研究显示:阿里红多糖水溶液可降低衰老小鼠体内MDA水平,提高SOD、GSK-PX活力,提示其可能有抗氧化和抗衰老作用;阿里红多糖水溶液能增加衰老小鼠免疫器官重量,具有一定的免疫增强作用。

英文摘要: Fomes officinalis Ames is a kind of traditional Uygur medicine and the polysaccharide is one of the effective constituents from it. This topic extracted the polysaccharide from Fomes officinalis Ames(Fop)and analyzed its basic structure. The contents of reducing sugar and uronic acids in Fop were 73.81 μ/ml and 17.07%,respectively. Fop was mainly composed of rhamnose, arabinose, xylose, mannose, glucose, and galactose with a molar ratio of 0.2:0.29:0.41:0.77:0.70:1.16. Meanwhile, Fop could be absorbed through intestinal epithelial cells and its intake and transport across Caco-2 cell monolayers were probably passive diffusion as the dominating process. On the intervention trial of the expression on the intestine mucosa lymphocytokines, Fop could decrease the amount of intestine mucosa lymphocyte and the level of TNF-α12289;IL-1βincrease the level of TGF-βwhich suggested that Fops might regulate the intestine mucosa immunity through decrease the activity of the local mucosa lymphocyte and the expression of the inflammatory factors.The results of pharmacodynamics experiments showed that the solution of Fop could increase the activities of SOD and GSH- PX, decrease MDA content in aged rats, which suggested that Fop might have the effects of anti- oxidation and anti-aging. Another experiment result showed that the solution of Fop could increase the immune organ weight of the aged rats and had certain immunopotentiation.
