中文摘要: 当前各种组学方兴未艾,其共同点都是期望从整体层面研究特定目标物质群(组)的整体组成及 相互关系,但是如何实现检测信息的全面性(整体)和高通量(快速)的同时兼顾仍是技术瓶颈。本项目基于化合物群分类检测的策略,探索建立一种分类标记扫描质谱法,即针对不同化合物群(组)的特点(如有机酸、氨基酸、生物碱、糖及其衍生物、同系物等),通过选择和优化合适的化学反应,使同一大类化合物(不同于特异性靶标标记)都接上一个相同的化学标签,不同类可以接上不同的标签,进而选择相应的质谱扫描方式分别获得各类化合物群的轮廓谱信息。相对于一般的中性丢失等扫描方式,该方法检测的化合物群范围大大扩大,"丢失"化合物信息少,信噪比提高;相对于全扫描模式,该方法过滤了本底噪声和复杂基质信号的干扰,因此可以对复杂样品按目标化合物群快速分类筛查。将该方法应用于临床与中药代谢组学研究,以期实现疾病标志物组合或中药活性成分群的快速筛查。

英文摘要: The emerging omics approaches had a common damand on the comprehensive characterization of all the components and their relationships among the ome while the bottleneck lay on the technologies of integrative and highthrough information acquisition. The proposal is to establish a group-label scaning mass spectrometric approach for grouping-based screening. It requires to label a characteristic tag to interested compound group (not a singe target compound) and then select a scaning mode of MS to obtain a profile of such a group of compounds.In comparison with the conventional scaning detection such as neutral lost scan, the proposal approach can extend the species of screening with less omitter of compounds and higher signal to noice. And in comparison with the conventional detection such as full scan mode, it can filtrate the noice and matrix signals and will be able to apply in the grouping-based screening of complex samples. The application of this approach in the metabonomic study of Traditional Chinese Medicine and desease screening will be also included in the project.
