中文摘要: 药物研发过程中的一个重要环节是安全性评估,利用小动物检查其对心肝肾等重要器官的毒副作用,这种安全性评估方法成本高且效率低。“人体器官芯片”则可以克服这些缺点,极大地简化了药物安全性评估过程。心肌组织芯片是“人体芯片”中的最核心部分,可以用于测试药物对心脏的毒副作用或功效,并且可以研究心脏组织的生理与病理过程。本项目针对目前心肌组织芯片构建中的人源心肌细胞成熟化和心肌组织立体结构这两个最突出的瓶颈问题,发展人源心肌细胞成熟化的培养技术,研究模拟心肌组织立体结构及心肌组织构建的方法,探索构建新型心脏组织芯片的技术。研究结果将为心脏芯片的构建提供新的思路,为获得低成本高效评估药物心脏毒副作用平台打下基础。该研究将推动人体芯片的发展,具有广阔的应用前景。

英文摘要: One of the key processes in drug development is evaluation of safety, using small animal to test the drug’s toxicity on the critical organs such as heart, liver and kidney. This kind of evaluation of safety is high cost but low efficacy. These shortcomings can be overcome by ‘organ-on-a-chip’ that facilitate the evaluation of safety. Myocardium-on-a-chip is the most essential component of ‘human-on-a-chip’ and can be applied to test cardiac toxicity of drugs. Meanwhile, myocardial chip can be used to study cardiac physiology and pathology. This study is aiming to solve the two key issues of myocardial chip: maturation of human cardiomyocytes and 3D myocardial structure. For these purposes, we will develop culture techniques to promote the maturation of human cardiomyocytes and to construct 3D myocardial structure in order to obtain novel myocardial chip. Results from this study will provide new method for constructing myocardial chip and set up a foundation for low-cost and high efficient platform to test cardiac toxicity of drugs. This study will promote the development of human-on-a-chip and is promising for future application.
