中文摘要: 我国是茶树的原产地,拥有茶组植物的全部自然种类,云南是茶及其近缘植物的分化中心。茶组植物中不仅存在着十分丰富的化学多样性,而且蕴藏着有意义的抗病毒活性物质。美国FDA批准的首个上市植物药即为茶多酚(Veregen),用于治疗人类乳突病毒引起的尖锐湿疣。本项目拟在前期分析研究的基础上,采用半制备HPLC和LC-MS联用检测技术,应用制备和半制备HPLC并结合传统方法,系统研究4-6种重要茶组植物的化学成分,特别是其中的特殊儿茶素类、黄酮类、水解单宁类、以及皂苷类成分等,揭示茶组植物的化学多样性,为茶组植物的系统分类提供植物化学方面的依据;同时,主要以DNA病毒单纯疱疹病毒I型和II型以及RNA病毒A型流感病毒 (IAV)为模式病毒,对分离到的化合物进行抗病毒活性测试,探讨构效关系,以期发现结构新颖的、有进一步开发潜能的抗病毒活性先导化合物,为茶组植物的合理开发利用和保护提供科学依据。

英文摘要: China possesses all the natural species of the Genus Camellia Section Thea, and is the origin of tea. In which, Yunnan is the distributing center of tea and its related species. Previous study suggested that there is not only the chemical diversity, but also potential antiviral substances in the species of the section Thea. Veregen, the first botanical drug product approved by FDA, is composing of green tea catechins used for the treatment of external genital and perianal warts caused by human papilloma virus (HPV). Guided by the combination of semi-preparative HPLC and LC-MS analysis, this project is going to study the chemical constituents, particularly the special derivated catechins, flavonoids, hydrolysable tannins and saponins, of 4 to 6 species from the Genus Camellia Section Thea, mainly by semi-preparative and preparative HPLC together with the traditional method for phytochemical investigation. It will not only reveal the chemical diversity of the species of Genus Camellia Section Thea, but also provide chemical evidence for the taxonomy and phylogeny of Section Thea. At the sametime, the isolated compounds will be evaluated for their antiviral activities against HSV-1, HSV-2 and IAV. Their structure-activity relationships will be discussed, in order to find novel and potential antiviral drug leads from the species of Section Thea based on further in vivo test. It will provide scientific evidence for the sustainable utilization and protection of section Thea.
