中文摘要: 荧光显微成像技术已成为活体组织与细胞的微观结构与形态可视化研究必不可少的重要手段,在肿瘤诊断和医疗检测等领域得到了广泛应用。小分子荧光探针对活体组织与细胞的特异性标记发挥着十分重要的作用。然而,目前对细胞器特异性标记及荧光增强的分子机制的认识不足严重阻碍了特异性荧光探针分子的设计合成及其在高分辨活细胞荧光显微成像技术中的应用。本项目立足于生物光子学领域的研究热点-探针标记和荧光显微成像技术,通过花菁类特异性小分子荧光探针分子设计、海拉活细胞标记与荧光显微成像、探针分子与细胞中生物大分子的结构与结合机制及光学效应与特性等方面的研究,阐明探针分子在活细胞选择性标记中的荧光增强分子机制。本项目具有生物光子学、化学、分子生物学等多学科交叉融合的特点及显著的创新性。通过本项目的研究工作,将为特异性小分子荧光探针的分子设计及其在分子水平的活细胞标记与高分辨光学成像技术的发展提供重要的科学基础。

英文摘要: Fluorescence microscopy imaging of living tissue and cells has been an essential method in visualizing the subcellular microstructures, which has attracted much attention in tumor research and medical diagnosis due to the increasing requirement of biophotonics at molecular level. Small molecule fluorescent probes have played an important role in labeling living tissue and cells in microscopy imaging. However, the application of the small molecule fluorescent probes with specific labeling property in high resolution fluorescence microscopy imaging of living cells has been hindered due to the insufficient understanding of the specific labeling of cellular organelle and the mechanism of the fluorescence enhancement. In this project, the applicant will focus on the attractive research filed in biophotonics, the fluorescent probe labeling and fluorescence microscopy. The molecular mechanism of fluorescence enhancement will be illustrated by developing a series of carbazole-based cyanine probes with specific labeling property, investigating specific labeling of living Hela cells and fluorescence imaging, demonstrating the structure, molecular interaction, optical effect and optical property of probes and cell organelles.
