中文摘要: 设计与合成高靶向、高灵敏、高疗效、高稳定和低毒副作用的"诊疗一体化"探针是治疗诊断学技术研究的关键内容之一。本项目拟结合纳米材料制备与修饰技术、分子工程技术以及活体荧光成像技术等,系统开展同时具备信号发生和光热治疗性能的金纳米材料制备与修饰研究以及具有复杂生理环境中靶肿瘤细胞激活性能的核酸适配体探针设计研究,并在此基础上,选取循环肿瘤细胞和活体肿瘤作为研究对象,构建一系列基于核酸适配体-金纳米材料功能组装体的新型"诊疗一体化"探针,发展血液中循环肿瘤细胞的高灵敏比色分析与选择性热疗技术平台和活体肿瘤激活式荧光成像及其引导下的靶向热疗技术平台,为实现肿瘤早期诊断与治疗提供新方法和新技术。这将是一项在分子工程技术和生物纳米技术等前沿交叉领域推动现代分析化学发展的创新性课题。

英文摘要: One of the key contents for theranostics is the design and synthesis of theranostic probes with high specificity, high sensitivity, high efficacy, high stability and low toxic side effects. In this project, by combining the technologies of nanomaterial preparation and modification, molecular engineering and in vivo fluorescence imaging, systematic studies will be performed on the preparation and modification of gold nanomaterials with both signal generating and photothermal therapy (PTT) functions, and the design of aptamer probes with activatable ability by target tumor cells in complex physiological circumstances. On this basis, by choosing circulating cancer cell (CTC) and tumor in living body as study objects, a series of novel theranostic probes will be constructed based on aptamer-gold nanomaterial functionalized assemblies. Moreover, two technology platforms will be developed, including the platform for the highly sensitive colorimetric analysis and selective PTT of CTC in blood and the platform for the in vivo activatable fluorescence image-guided tumor PTT.
