中文摘要: 次级毛囊的周期性再生是影响羊绒产量和质量的关键因素,目前绒山羊次级毛囊周期性再生的分子调控机制尚不清楚。人和小鼠的研究表明,毛囊周期性再生受生物钟基因节律性表达的控制,而生物节律性与表观遗传修饰密切相关。本项目拟以绒山羊次级毛囊毛乳头细胞(DPCs)为模型,研究生长期、退行期和休止期次级毛囊及DPCs中钟基因的表达规律,确定影响次级毛囊再生的关键钟基因;测定不同时期关键钟基因启动子区组蛋白乙酰化程度,分析表观遗传修饰对次级毛囊周期性再生的作用;利用CHIP-PCR及RNAi技术分析CLOCK:BMAL1对关键钟基因表达的影响,及其与关键钟基因启动子区组蛋白修饰间的关系。从而揭示生物钟基因调控绒山羊次级毛囊周期性再生的作用机制,为绒山羊生产中改变羊绒的生长周期,提高羊绒产量和质量提供科学理论依据。

英文摘要: Periodic regeneration of secondary hair follicle is a key factor affecting the quantity and quality of goat cashmere. But currently, its molecular mechanism is still unclear. It had been shown that regeneration cycle of hair follicle is regulated by rhythmic expression of circadian clock gene according to the results of mouse and human, and biological rhythm is closely related to epigenetic modification. In this project, dental papilla cells (DPCs) are acted as a model. In order to find the key clock gene, we will anlyze the expression characteristics of clock genes in secondary hair follicles and DPCs from anagen, catagen and telogen. Then, we will examine histone acetylation level of promoter region of key clock gene in different development stage to illuminate the role of epigenetic modification in periodic regeneration of secondary hair follicle. Finally, we will anlyze the effect of CLOCK:BMAL1 on the expression of key clock gene and the relation between CLOCK:BMAL1 and histone acetylation of promoter region in key clock gene using CHIP-PCR and RNAi. All these will be benefit to reveal the mechanism of periodic regeneration of secondary hair follicle in view of epigenetic and provide theoretical basis for improving the quantity and quality of cashmere by changing the periodicity of cashmere growth.
