中文摘要: 本研究在大量前期工作的基础上,对无功能性胰腺神经内分泌肿瘤(NF-PNET)患者与正常人外周血中的miRNA表达进行检测,通过探索NF-PNET发病特征性miRNA的表达,可在基因转录后水平阐释其发病机理;获得的NF-PNET miRNA“指纹图谱”,可用于疾病的诊断与鉴别诊断,亦可作为其治疗的潜在靶点;同时,本研究借鉴2014年中国学者张辰宇发表在《cell research》上的突破性研究成果,率先从清胰化积方外源性miRNA“跨物种调节”的角度探索中药复方的作用机理,阐释对NF-PNET有良好疗效的清胰化积方在基因转录后水平的作用机理,并寻找其作用的靶基因及靶信使RNA;从“跨物种调节”的角度探索中医药的疗效机理,可为中医药治疗肿瘤的研究提供崭新的思路,并为清胰化积方治疗NF-PNET提供充分且有说服力的实验室依据。

英文摘要: Based on our former work, this study is going to detect the miRNA expression in non-functional pancreatic neuroendocrine tumor(NF-PNET) and healthy controls. By exploring the characteristic miRNA expression of NF-PNET, its pathogenesis on post-transcriptional level may be discovered. The acquired fingerprint spectrum can be used for its diagnosis and differential diagnosis,as well as the promising therapeutic targets; Plus, according to the scientific breakthrough published in <cell research> ,2014, by Chinese scientists,this study at the first time explore the mechanism that exogenous miRNA from herbs of Qingyihuaji Recipe intervene the endogenous miRNA transcription. Thus, the gene terget of Qingyihuaji Recipe’s may be discovered. This study will also provide persuasive evidence from laboratory to support the good therapeutic response with Qingyihuaji Recipe. Also, this study may give a brand new idea in scientific research of Chinese medicine.
