中文摘要: 异基因造血干细胞移植是根治恶性血液病的最有效方法,但由于MHC屏障,存在移植物抗宿主病(GVHD)发生率高、免疫重建慢等问题。近年来的研究发现,间充质干细胞(MSCs)具有非特异性的免疫抑制和抗增殖作用。加入供者骨髓MSCs的混合移植能够有效地防治GVHD、诱导免疫耐受,但对MSCs作用机制看法不一。而Dickkopf-1(DKK-1)分子是一个新进热点研究的内源性分泌因子,参与细胞的增殖、分化等过程。我们预实验结果显示,抑制MSCs 细胞DKK-1的表达,能够抵消MSCs对T细胞增殖的抑制作用。因此,本实验拟从细胞及分子水平,采用RNAi和真核表达质粒转染技术,抑制或过表达DKK-1后,通过体外共培养等方法研究骨髓MSCs对受者细胞毒T细胞、Treg、DC等细胞调节作用,阐明DKK-1分子在MSC诱导免疫耐受中的作用机制,为提高移植效率、预防GVHD和免疫功能重建奠定理论基础。

英文摘要: Allergenic hematopoietic stem cell transplantation is an effective treatment for malignant hematopoietic disorders, but it exists a problem of graft versus host disease and needs a long time to re-establish self-tolerance due to MHC barrier. Recent studies have shown that MSCs could improve the non-specific immune suppression and antiproliferation role. Although mixed transplantations of donor MSCs and PBSC can prevent GVHD and induce immunologic tolerance, opinions differ on its mechanism of regulatory effects.Dickkopf-1(DKK-1)as a new endocrine factor has become a hot issues and is associated with the functions of cell proliferation and secretion. Our preliminary findings demonstrated that decreased expression of DKK-1 in MSCs could compensate for the inhibitory effect in T lymphocyte. Therefore, to validate the functions of the potential MSCs,we investigate the effects of immunity regulation of DKK-1 toward receptor's DC、Treg at molecular, cellular and animal model levels by co-culture method, RNAi and overexpreesion system. The Eluciating Mechanism of DKK-1 regulatory effects on MSC-induced Immunologic tolerance, will open up an new way for the prevention of GVHD and re-establish self-tolerance.
