中文摘要: 血管平滑肌细胞增殖在糖尿病动脉粥样硬化斑块形成中至关重要,但机制尚不完全清楚。病理条件下,内皮细胞分泌功能改变会刺激平滑肌细胞增殖、促进粥样斑块形成。我们发现高糖环境下内皮细胞KLF2下调导致FGF5表达分泌增加,FGF5具有刺激细胞增殖作用。本课题以内皮细胞和平滑肌细胞的cross talk为切入点,通过建立内皮细胞-平滑肌细胞共培养的细胞模型,ApoE-/-小鼠给予高脂饮食并腹腔注射STZ建立糖尿病动脉粥样硬化动物模型,借助KLF2过表达、FGF5干扰等方法观察高糖刺激下内皮细胞KLF2-FGF5表达变化,从分子、细胞和整体水平探讨糖尿病内皮细胞KLF2-FGF5改变对平滑肌细胞增殖对VSMC增殖和细胞内MAPK通路活性的影响,明确KLF2作为FGF5转录抑制子的地位,探讨该通路对糖尿病动脉粥样硬化斑块形成的作用。本研究的顺利开展,将为糖尿病大血管并发症的防治提供新思路和新靶点。

英文摘要: Atherosclerosis is a main complication in diabetes, acute cardiocerebral vascular disease caused by plaque rupture and thrombosis is the main reason of mortality in diabetes. Proliferation of vascular smooth muscle cells plays a pivotal role in atherosclerosis, however, the underlying mechanism is not clear yet. We proved that high glucose inhibited KLF2 expression in HUVECs, a critical transcription factor, and therefore induce FGF5 expression and secretion, which is potential in stimulate cell proliferation. In this study, we will try to illuminate the role of KLF2-FGF5 pathway in VSMC proliferation through EC-VSMC coculture,KLF2 over expression and FGF5 siRNA knockdown. Meanwhile, we will also establish diabetic atherosclerosis animal model by STZ infusing in ApoE-/- mice fed with high fat diet, and interfere FGF5 expression through shRNA lentivirus, and prove the role of KLF2-FGF5 in vivo. Our finding will help with the prevention and treatment of vascular complications in diabetes.
