中文摘要: 最新研究表明,p38MPAK信号通路与基质金属蛋白酶(MMPs)表达和活性改变密切相关。我们前期研究发现,一定参数的电磁脉冲(EMP)辐照可导致血脑屏障(BBB)紧密连接(TJ)蛋白ZO-1降解和MMPs表达及活性的增加,同时还观察到p38蛋白表达增加,因此我们推测:p38MPAK信号通路调控的MMPs表达和活性改变可能参与了EMP辐照致TJ 蛋白的降解过程。本项目拟通过研究EMP辐照后TJ蛋白ZO-1、MMPs、p38MPAK信号通路相关蛋白的变化,分析三者的变化规律及相关性,评价p38MPAK信号通路在EMP致MMPs增加中的作用,并通过p38 MPAK信号通路抑制剂的应用,明确p38 MPAK信号通路调控MMPs在EMP致BBB TJ蛋白降解中的作用,为EMP致BBB的损伤防护提供新思路。

英文摘要: Recent studies have shown that p38 MPAK signaling pathways are closely related to the change of matrix metalloproteinases (MMPs)expression and activity. Our initial findings revealed that the electromagnetic pulse (EMP) irradiation at special parameters can lead to the degradation of blood brain barrier tight junction (TJ) protein and the increase of MMPs expression and activity.Additionally, the increase of p38 protein expression was observed, so we speculated that p38 MAPK signaling pathways regulating the change of MMPs expression and activity may participate in the process of EMP irradiation resulting in the degradation of TJ protein.We evaluate that the change of TJ protein levels, MMPs activity , proteins involved in p38 MAPK signaling pathways after EMP radiation in order to analyze its correlation and evaluate the role of p38 MAPK signaling pathways in EMP leading to the increase of MMPs activity. In addition, we definite the role of p38 MAPK signaling pathways in EMP causing the damage of closely connected BBB through p38 MAPK signaling pathway inhibitor to provide a new way for the prevention of the injury of BBB caused by EMP radiation.
