中文摘要: 组蛋白去乙酰化酶(HDACs)介导的组蛋白修饰作为基因转录调控的重要方式而广泛参与到植物生长与发育途径。目前,对HDACs在基因转录调控中相互作用的转录因子、蛋白质复合体的组成及调控的靶基因网络的尚不清楚。本项目前期研究表明,拟南芥RPD3/HDA1型组蛋白去乙酰化酶HDA15与bHLH类转录因子PIF3相互作用,并共同参与抑制叶绿素的生物合成;HDA15与其它bHLH类蛋白PIF1和bHLH147也能发生互作。本项目拟在此基础上,继续筛选和验证与HDA15互作的bHLHs蛋白;分析HDA15与bHLHs互作共同调控的生物学过程;分析HDA15蛋白质复合体组成及调控的靶基因网络。以期揭示HDA15与bHLHs转录因子相互作用的生物学功能,阐明植物生长与发育过程中的表观遗传调控机制。

英文摘要: Histone deacetylases play an important role in regulation of gene expression during plant growth and development. Currently, the interacting proteins, component and direct target genes of HDACs are still unknown. In our previous studies, we showed that HDA15, an RPD3/HDA1-type HDAC, interacts with bHLH protein PIF3 in repression of chlorophyll biosynthesis in Arabidopsis. Furthermore, HDA15 can also interact with PIF1 and bHLH147. In this study, we will go on screening and confirming the HDA15-interacting bHLH proteins. Next, we will study the development pathways co-regulated by HDA15 and bHLHs. Furthermore, we will study the component of HDA15 protein complexes and detect direct target genes of HDA15 in Arabidopsis. Based on the work above, we will try to clarify the biological function mediated by HDA15-bHLHs interaction and the epigenetic mechanisms in regulation of plant growth and development.
