中文摘要: 响应信号的强度与检测目标物丰度反相关的金属薄膜增强/淬灭荧光型微孔板芯片是一种能更大限度的提高检测灵敏度的新型微阵列芯片。食品中痕量重金属离子的污染已经严重影响到人们的身体健康。本项目拟构建多元,高灵敏检测食品中重金属的新方法,探讨实现金属薄膜增强/淬灭荧光型,响应信号的强度与检测目标物丰度反相关的微阵列芯片的具体条件。具体而言,在载玻片表面自组装上金纳米颗粒薄膜,以此作为荧光信号的增强与淬灭源;用石墨烯量子点标记脱氧核酶,将脱氧核酶与键合于芯片基底表面的脱氧核糖核酸底物链杂交,形成金属薄膜增强/淬灭荧光型微阵列芯片;以多种重金属离子的同时检测为具体实施内容,运用脱氧核酶特异识别重金属离子时,结构发生卷曲的性质来调节石墨烯量子点与金属薄膜的距离,从而淬灭石墨烯量子点的荧光,利用响应信号的强度与检测目标物丰度反相关的关系,实现对重金属离子的多元,高灵敏检测。

英文摘要: Metal film-enhanced/quenched fluorescence micro-plate arrays, which the response signal intensity is inversely correlated with the abundance of detection target, is a novel microarray, that is capable of greatly improving the detection sensitivity. The contamination of trace amount heavy metal ions in foods has already greatly influenced the people's health. This project intends to construct a multiplex, high sensitivity method to determination of heavy metal ions in foods, and to discuss the realization of the actual condition to novel microarray. In practical terms, utilizing the Au-nanoparticle self-assembly to dispose the glass surface, forming the source of enhanced and quenched fluorescence;using graphene quantum dots labeled with DNAzymes and hybridizing DNAzymes with the complementary DNA that combining to the chip surface, forming the metal film-enhanced fluorescence microarray. For simultaneous detect multiple heavy metal ions as the concrete implementation content,using the DNAzymes capability of high efficiency and sensitivity identification of the heavy metal ions, applying the heavy metal ions quenching effect to graphene quantum dots, utilizing the anti-correlation between the response signal intensity and the abundance of detection target, realized the multiplex, super-high sensitivity determinat
