中文摘要: 鱼类早期发育阶段个体发育形态特征变化剧烈,骨骼生长发育是个体发育的重要环节,研究鱼类早期骨骼发生、发育特征及其调控机制,对揭示鱼类早期发育阶段的功能趋向及环境适应具有重要理论意义和实践价值。脊椎动物骨骼形成多受骨形态发生蛋白(BMP)调控,其中BMP2、BMP4又是诱导成骨活性最强的因子,研究它们的遗传学特征及功能是从分子水平认识骨骼生长发育的基础。半滑舌鳎(Cynoglossus semilaevis)是我国重要海水增养殖对象,目前国内外有关其骨骼发生、发育调节机制研究尚属空白。本项申请拟在系统观察半滑舌鳎早期骨骼发生、发育特征基础上,研究BMP2和BMP4的基因结构特征和时空表达模式,从形态和分子两个水平探讨BMP相关基因与早期发育阶段骨骼发生、发育的相关性,探讨半滑舌鳎早期骨骼发生、发育的调控机制,丰富硬骨鱼类早期发育生物学研究,为优化与创新养殖鱼类苗种生产技术提供理论依据。

英文摘要: Ontogenetic morphological characteristics of fish change dramatically at early life stages. Moreover, growth and development of skeleton is important in the ontogenetic process. Consequently, clarifying the skeletal development characteristics and their regulation mechanisms at early life stages are of theoretical significance and practical value for explanation of the fish functional trends and environmental adaptation. The vertebrate bone formation is regulated by bone morphogenetic proteins (BMP), among which BMP2 and BMP4 exhibit the highest osteoinductive activities. Investigating the genetic characteristics and function of these factors may help understanding the skeletal development at the molecular level. The Tongue sole (Cynoglossus semilaevis) is regarded as a perfect target species for sea farming and sea ranching. To date, the regulatory mechanisms of skeletal development in C. semilaevis has thus far only rarely been reported. In this study, on the basis of the systematic observation of skeletal development characteristics of C. semilaevis, genomic structure and mRNA expression (temporal and spatial expression pattern) analysis of the BMP genes (BMP2 and BMP4) will be performed in order to examine the association between the BMP genes and early skeletal development at both the morphologic and molecu
