中文摘要: 基因在不同倍性的植株中会发生激活或沉默现象,可能导致新性状出现或消失。了解植物不同倍性植株的基因表达差异以及是如何影响表型性状的,对于竹子遗传改良极具重要意义。项目组在麻竹花药培养同一愈伤组织继代过程中获得了三倍体(2n=3X=36)、六倍体(2n=6X=72)和十二倍体(2n=12X=144)三种倍性的再生植株,通过表型观测发现,不同倍性植株间的出笋量、气孔大小、叶绿素含量、光合、蒸腾速率等性状存在显著差异。本研究将以这三种遗传背景一致的不同倍性植株以及杂交F1代的叶片为材料,利用新一代测序(RNA-Seq)技术分析基因在不同倍性植株间的表达差异,通过qRT-PCR和Northern杂交技术验证差异基因的表达模式,同时结合表型性状,探讨不同倍性麻竹植株性状差异的分子调控机理,分离克隆2-3个控制麻竹重要经济性状的功能基因,为麻竹分子遗传育种和功能基因组学研究奠定基础。

英文摘要: Gene in the different ploidy plants will activate or silence, could lead to a new character appears or disappears. Understanding the difference of gene expression in different ploidy plants and how it affects the phenotypic traits is extremely important for genetic improvement of bamboo. Triploid (2n=3X=36)、hexaploid (2n=6X=72) and dodecaploid (2n=12X=144) were obtained from the same origin of embryogenic callus of Dendrocalamus latiflorus Munro by anther culture. Found through phenotypic observation, shoot yield, stomatal size, chlorophyll content, photosynthesis and transpiration rate of the different ploidy plants character significant difference. Leaves transcriptome profile of different ploidy anther-regenerated plants and F1 seedling samples will be compared using next-generation sequencing (RNA-Seq) technology. Gene expression patterns will be confirmed by qRT-PCR and Northern hybrid technology. While combining with pheotypic traits, we will explore the molecular mechanism of plant characteristics difference in different ploidy plants. Finally, 2-3 functional genes controlling important economic characters will be isolated and cloned. Our study will lay a certain foundation to functional genomics research and genetic breeding of bamboo.
