中文摘要: 重金属对生态系统造成的污染已成为全球关注的问题,我们前期研究表明藻类能吸附水体中重金属,重金属也能对藻类产生一系列的毒理效应。为了更深入了解藻类对重金属耐性和解毒机理,本项目拟以一种大型褐藻-巨藻和六价铬为研究对象,首先采用x射线能谱分析仪、差速离心以及逐步化学提取法研究了铬在巨藻组织和亚细胞水平上的分布特点和化合形态特征;其次以同位素相对绝对定量技术(iTRAQ)为基础,采用联合多维固/液相色谱技术与串联质谱技术分析铬胁迫下蛋白质的变化,并通过蛋白质印迹法进行验证;最后采用气相色谱-质谱联用(GC-MS)技术和核磁共振(NMR)技术分析铬胁迫下代谢产物变化。本研究的完成可以从亚细胞水平揭示铬离子在巨藻内的迁移、转化以及参与重金属铬解毒的可能途径,鉴定到的铬胁迫响应蛋白和代谢产物将为阐明巨藻对铬胁迫响应的分子机理提供理论依据。

英文摘要: Heavy metal pollution of ecosystems is a global problem. Our previous research showed that algae can adsorb the heavy metal and result in a series of toxicological effect. In order to further understand the mechanism of heavy metal detoxification and tolerance in alga. This project intent to focus on the brown alga- Durvilaea potatorum and chromium (Ⅵ). Firstly, the characters on subcellular distribution and chemical forms of chromium are acquired by X ray energy dispersive spectrometer, differential centrifugation and gradually chemical extraction method. Secondly, iTRAQ is combined with two-dimensional liquid chromatography and MS to identify the differentially expressed protein under Cr stress, and Verified by Western blotting. Thirdly, the change in metabolic productions is analyzed by the nuclear magnetic resonance (NMR) and gas chromatography-mass spectrometry (GC-MS) under Cr stress. This study can reveal Cr migration, transformation and Cr detoxification in Durvilaea potatorum. The differentially expressed proteins and metabolic productions can afford a theoretical evidence for further clarifying Cr tolerance molecular mechanism.
