中文摘要: 对甲酚是一种高毒的芳香化合物,作为重要的化工原料在使用过程中大量地泄漏到环境中,严重污染环境并威胁人类健康。微生物降解对甲酚的研究主要集中于革兰氏阴性菌假单胞菌中,而在革兰氏阳性菌中鲜见报道。此外,目前的研究局限于酶的分离及中间产物的鉴定,而酶的编码基因则很少被克隆到,并且其调控机制也不清楚。谷氨酸棒杆菌是一株革兰氏阳性好氧菌,可降解包括对甲酚在内的多种芳香化合物。本项目拟结合比较蛋白质组学和基因敲除/互补方法鉴定谷氨酸棒杆菌中的对甲酚降解基因簇;利用生物化学和分子生物学技术分离对甲酚降解酶并鉴定中间代谢产物,确定降解酶的编码基因;并利用转录调控分析方法对降解途径中的调控机制进行分析,以从分子水平上揭示谷氨酸棒杆菌降解对甲酚的代谢途径和调控机制。本项目对于认知革兰氏阳性菌中对甲酚降解的分子机理具有重要意义。

英文摘要: As an important industrial raw material, p-cresol is a main contaminant with high toxicity in the environment and threats human health. The research of microbial degradation of p-cresol is focused on Gram-negative Pseudomonas and limited in enzyme isolation and intermediate product identification. However, the gene cluster of p-cresol degradation is seldom reported. Additionally, little is known about p-cresol degradation in Gram-positive bacterium. Corynebacterium glutamicum, a Gram-positive aerobic bacterium, can degrade a variety of aromatic compounds including p-cresol. In this research, the gene cluster of p-cresol degradation in C. glutamicum will be identified via comparative proteomics and gene knockout/complementation approach. Using biochemical and molecular biological approach, the enzymes and intermediate products in p-cresol degradation pathway will be isolated and identified. Furthermore, the regulatory mechanism of p-cresol catabolism will be analyzed. Our aim is to reveal the catabolic pathway and regulatory mechanism of p-cresol degradation in C. glutamicum. This research is helpful to understand the molecular mechanism of p-cresol degradation in Gram-positive bacterium.
