中文摘要: 中枢orexin能神经系统和中枢组胺能神经系统均特异性地起源于自主活动调控的高级中枢下丘脑,且都发挥了全脑功能调节者的作用。然而,目前对它们在机体运动调控这一脑重要功能中的作用仍知之甚少。我们新近的工作首次揭示了orexin在经前庭神经核介导的运动调控和在重大运动疾病猝倒症中的作用(Zhang et al: Neuron, 2011),也初步揭示了组胺在中枢运动调控中的作用。本项目拟在我们这些系列研究工作的基础上,从整合生理学的角度出发,在分子、细胞、神经环路和整体动物行为水平上,深入和系统地研究中枢orexin能和组胺能神经系统对皮层下重要运动结构(小脑、基底神经节、前庭核和脊髓)和经这些结构介导的运动调控、运动学习和运动疾病的作用和机制。研究结果不仅具有重要的神经生物学基础理论意义,而且对于临床深入认识和治疗重大运动疾病(帕金森病、共济失调和猝倒症等)具有重要的指导价值。

英文摘要: Central orexinergic system and histaminergic system both originate from the hypothalamus, a high center for autonomic regulation, and exert a general modulator for whole brain activity. However, role of these two systems in motor control still remains enigmatic. Our recent study has revealed, in the first time, a role of orexin in central vestibular motor control and cataplexy, a severe motor disease (Zhang et al: Neuron, 2011). We also demonstrate function of histamine in central motor control. Therefore, on integrative physiological viewpoint and at molecular, neuronal, circuit, and behavioral levels, the present study will focus on the role of central orexinergic and histaminergic systems in important subcortical motor structures such as the cerebellum, basal ganglia, vestibular nuclei and spinal cord. Functions of orexin and histamine on motor control, motor learning and motor diseases will be especially investigated. The results will not only are of important and general significance in neurobiological theory, but also contribute to develop novel clinical strategies and new targets for treatment of motor diseases including Parkinson’s disease, ataxia and cataplexy.
