中文摘要: 菌核病是油菜的主要病害,到目前为止尚未在油菜及相关植物中找到抗性基因。油菜等植物中的PGIP蛋白对核盘菌的致病因子PG具有诱导性防卫反应作用,有关PGIP蛋白的防卫反应机制的研究已成为热点。本研究应用同源序列法,分别从甘蓝型油菜抗、感病材料中克隆全部17个已报道的BnPGIP基因,在DNA水平上比对分析抗、感病材料中BnPGIP基因的SNP位点差异;利用荧光定量RT-PCR筛选与抗性相关的候选BnPGIP基因,并对候选基因进行克隆,应用转基因技术对克隆的PGIP基因进行功能分析。研究结果能为揭示油菜中内源BnPGIP基因的抗病机制和功能提供理论基础,同时也为通过基因工程技术应用内源BnPGIP基因提高油菜的抗菌核病能力,为油菜及相关植物抗菌核病育种开辟新的途径。

英文摘要: Sclerotinia is the main disease of rape. Genes involved in defending Sclerotinia sclerotinia have not been found in rape and some other related plants so far. Some researches revealed that PG protein from sclerotinia sclerotiorum is the key pathogenic factor. PGIP protein exists in some plants is proved as an inhibiting factor targeted to PG protein and induce the defense reaction ability of plants. The research about defense reaction mechanism of PGIP protein has become the focus right now. This study is to clone all the 17 PGIP genes reported in Brassica Napus L. from resistive and susceptive rape material respectively, using homologous sequence method. Then check out the BnPGIP gene's SNP sites between resistive and susceptive rape material through DNA blast. We want to use fluorescence quantitative PCR to analyse the expression profile of 17 PGIP genes and select the candidate genes which may have function in defending sclerotinia. Then we clone the candidate genes' full length of cDNA from resistive rape and construct the overexpress vector, then express the PGIP genes in Arabidopsis and susceptive rape to confirm the function of candidate genes through transgenic technology.
