中文摘要: 本项目利用生物信息学、分子生物学等方法,成功构建了阿魏酸酯酶cDNA文库;获得了特定环境微生物的阿魏酸酯酶基因库,进行了阿魏酸酯酶基因的克隆、表达和性质测定;阿魏酸酯酶水解植物细胞壁条件的确定;阿魏酸酯酶与纤维素酶协同作用的单因素实验和最佳条件优化等方面的工作,使还原糖产量提高5%左右,这对生物质能源的利用具有重要的现实意义,建立了利用气相色谱快速、准确测定阿魏酸的方法,节约了测定成本。

英文摘要: The cDNA library of ferulic acid esterase was successfully constructed by the methods of bioinformatics and molecular.We had obtained environmental microbial ferulic acid esterase gene pool form specific environmental.The gene of ferulic acid esterase had been cloning and expression. The character of enzyme had been determined. Study on the condition of ferulic acid esterase hydrolyzed plant cell wall and the work of synergistic aspects of single factor and the optimum conditions with ferulic acid esterase and cellulase.The yield of reducing sugar increased about 5%. It was important practical significance for the use of biomass energy.The method of rapid and accurate was established to determined ferulic acid by gas chromatography. The determination of cost was saved with this method
