中文摘要: 本项目拟首先通过形态分析、分子鉴定和化学分析等手段确定200份香菇样品中的产毒菌株和脱氧雪腐镰刀菌烯醇(DON)及其隐蔽型毒素的暴露种类与含量水平,获得香菇中易污染的真菌种类及其产毒能力等基础信息。然后,将产毒菌株人工接种至香菇基质,在不同因素条件(温度、水活度、培养时间等)下培养,提取RNA,构建同源基因进化树,考察在DON及隐蔽型DON生物合成过程中起作用的关键基因,并研究主要因素对产毒基因蔟表达量的影响规律;提取香菇样品,考察不同因素下DON及隐蔽型DON的种类及表达量,建立次级代谢图谱,探明与DON及其隐蔽型的产生密切相关的代谢小分子,比对分析分子鉴定和化学分析结果,揭示香菇中DON和隐蔽型DON的发生规律及其特异性产毒基因蔟调控机制,为开展香菇中真菌毒素污染的风险评估、完善香菇质量安全控制技术标准提供理论依据及实验基础。

英文摘要: In this current research proposal, the fungal strains, types and concentrations of deoxynivalenol (DON) and its masked forms from a total of 200 Lentinula edodes are firstly evaluated by morphological identification, molecular characterization and chemical analysis to obtain the the basic information about the toxigenic types and levels of the seperated strains. Via artificial inoculation of the strains to Lentinula edodes under various factors (temperature, water activity,cultivation time), RNA is extracted and the homologous gene phylogenetic is established to identify the key gene clusters that play important roles on the biosynthsis of the DON and its masked forms. The regulation effects of the conditions on the specific gene clusters are also investigated. On the other hand,after extraction of the Lentinula edodes, the types and contamination levels of DON, masked DON and the related metabolites corresponding to the production of DON and masked DON under different conditions are clearly identified and quantified to develop the metabolic figures. After comparative analysis of the molecular identification and chemical analysis results, the production of DON and its masked forms and the regulation mechanism of the specific mycotoxin-producing gene clusters in Lentinula edodes are revealed to provide the theore
