中文摘要: 相位板成像技术是提高透射电镜相位成像衬度的重要技术方法之一。主要应用于冷冻生物样品结构解析。成像分辨率的损失限制了该技术在生物大分子结构解析的应用。本项目通过研制独立提出的可原位微区加热的半覆盖式薄膜相位板,拟解决目前的技术瓶颈。研究内容包括:1)可原位微区加热的半覆盖式薄膜相位板的研制及其评价;2)适用于Titan Krios透射电镜的高精度相位板系统的研制;3)使用研制的新型相位板系统对生物大分子进行成像和高分辨率结构解析。期望通过本项目的实施,推进相位板技术在生物大分子高分辨率结构解析领域的应用,并为球差校正正技术应用于生命科学领域奠定基础。

英文摘要: Phase plate imaging technique is one of the important technical method to improve transmission electron microscopy phase contrast imaging. This method used in hydrated frozen biological sample structure analysis. The imaging resolution loss limits the application of this technology in the analysis of the structure of biological macromolecules. For this project, a kind of part covered thin film phase plate with micro heater can be diveloped.And It can be expected to realize high resolution 3D reconstruction. Research contents include: 1) development and evaluation of this kind of part covered thin film phase plate with micro heater; 2) development of high precision positioning phase plate system equipped on Titan Krios transmission electron microscope; 3) realize the high resolution 3D reconstruction of biological macromolecules by using the new type phase plate system. Through the implementation of the project, the phase plate technology can be developped and applicated widely in the high resolution 3D reconstruction of the biological macromolecules,and applied to spherical aberration corrected microscope to use in the field of life science.
