中文摘要: HIV相关痴呆成为HAART时代研究的新难题,HIV在中枢神经系统的潜伏机制仍不是很清楚。不同的HIV亚型和不同组织来源的HIV序列存在差异,国内关于HIV相关脑病机制的研究报道较少。本研究拟分析神经中枢系统(CNS)与血液HIV LTR和Tat序列的多样性以及序列多样性与痴呆程度的相关性。本研究拟从转录的起始、延伸和表观遗传学方面,研究CNS HIV病毒在T细胞、巨噬细胞和胶质细胞的转录活性异同。利用针对Tat和NF-κB的抑制剂研究抑制剂对中国神经中枢系统来源的病毒转录的抑制程度。该研究将深化对中枢神经系统HIV的转录调控、HIV在中枢神经系统的潜伏机制的认识,将为抗病毒药物的研制提供重要的实验根据,为根除HIV病毒提供新思路。

英文摘要: AIDS related dementia is a concern in the Highly active antiretroviral therapy (HAART) era, and it is not clear about the viral latency mechanism in the central nervous system. The divergence and diversity of sequences exist in different HIV subtypes and in different tissues, currently, little information on HIV neuropathogenesis is available in Chinese settings. In the study, we will analyze the divergence and diversity of HIV long term repeat (LTR)AND Tat from paired paired cerebrospinal fluid (CSF) and plasma samples, and then evaluate the association between the sequences divergence and diversity and the stage of AIDS related dementia. We will investigate the HIV(CNS) transcription activity in T cell,macrophages,and Microglial cells, and the activity will be studied in the following aspects: transcription iniation, transcription elongation and epigenetics.The inhibitors targeted Tat and NF-κB will be used to evaluate their efficency in inhibiting HIV (CNS) transcription. The study will gain insight into the regulation of HIV gene transcription and the viral latency mechanism in the CNS, and the findings will provide valuble information on producing antiviral drugs and eradicating HIV in the CNS.
