中文摘要: 我们研究表明DST作为转录因子,通过调节Gn1a/OsCKX2基因的表达,影响花序分生组织中的细胞分裂素含量,从而控制水稻穗粒数的形成并最终影响产量。为了进一步研究DST的功能,我们通过酵母双杂筛选得到了与DST相互作用的蛋白DIP1。序列分析表明DIP1编码一个转录中介体亚基。转录中介体起到联系基因特异的转录因子与基于RNA聚合酶II的转录机器的作用,参与转录调控过程。拟南芥中的研究表明,中介体亚基通过与不同的转录因子相互作用参与到植物生长发育及激素信号转导的各个过程中;而目前对于水稻中介体亚基的报道还是空白。本研究拟从水稻中介体亚基DIP1基因入手,通过订购突变体,构建过表达及RNAi转基因植物,研究DIP1在产量形成上的作用;通过构建双突变体的方法研究DIP1与DST在遗传学上的关系;在分子水平上,进一步揭示二者如何协同作用并对OsCKX2等基因进行表达调控,从而控制水稻产量形成。

英文摘要: DST as a transcriptional regulator directly induced the expression of Gn1a/OsCKX2 (Grain number 1a/Cytokinin oxidase 2). The level of OsCKX2 influenced the cytokinin content in the shoot apical meristem which finally controls the rice grain number and yield formation. In order to discover the detailed function mechanism of DST, we screened the interaction proteins of DST through yeast two hybrid assays and got one candidate protein, DIP1. Sequence alignment revealed that DIP1 is the homologue protein of one Mediator subunit. Mediator complex is an evolutionarily conserved, multi-subunit complex that serves as a bridge between gene specific transcription factors and the RNA polymerase machinery to regulate transcription. In Arabidopsis, different Mediator subunits are involved in many development and hormone signaling pathways through interacting with different transcriptional regulators. And we know nothing about the rice Mediator subunit from existing study reports. In this study, we will focus on the function of DIP1 in the rice yield formation control through ordering the rice dip1 mutant and constructing the DIP1-overexpression and RNAi transgenic plants.
