中文摘要: 恶性肿瘤已成为严重危害人类健康的一组常见病、多发病,研究各种恶性肿瘤的发病原因,借助分析技术、纳米技术、生物技术和传感技术等学科的融合,找寻和发展新的分析检测方法,是实现恶性肿瘤早发现、早诊断、早治疗的重要发展方向。本项目以发展检测恶性肿瘤细胞及其相关物的新技术和新方法为目标,结合分析化学、材料科学、生命科学等交叉学科的前沿研究成果,采用超声化学、微波化学等方法合成如贵金属纳米簇、多元半导体、石墨烯、聚合物等新型量子点及其功能性复合材料,将其与白血病、乳腺癌等特定肿瘤细胞及其相关物的靶向生物分子相结合,组装具有识别功能的生物纳米探针,设计并构筑多尺度协同的生物传感界面,构建基于新型量子点检测白血病和乳腺癌的传感平台,利用光、电及相关技术对肿瘤细胞及其相关物进行高灵敏特异性检测,为实现恶性肿瘤的早期诊断、个体化药物治疗以及预后评估提供新途径。

英文摘要: Malignant tumor is a type of high incidence disease threatening human health. Thus it is essential to investigate the mechanism of malignant tumor and develop suitable analytical methods for their early diagnosis and treatment. Our aim in this project is to devise novel technologies and methodologies for the detection of malignant tumor cells and their correlates. Based on the progress of cutting-edge research in analytical chemistry, material science, and life science, novel quantum dots (QDs) (such as noble metal cluster, multicomponent semiconductor, graphene and polymer QDs) as well as their functional composites will be synthesized via sonochemistry, microwave-assisted method, etc. These nanomaterials will be further conjugated with target biomolecules for leukemia and breast cancer cells as well as their correlates, and assembled into bionanoprobes with the capability of specific recognition. By designing multi-scale synergetic biosensor interface, we intend to construct QD-based sensor platforms for detection of leukemia and breast cancer. Taking advantage of the optic and electric techniques, the obtained sensor platforms will be employed to the highly sensitive and specific detection of tumor cells and their correlates, and potentially provide new approaches for early diagnosis, personalized chemotherapy and prognosis evaluation to malignant tumor.
