中文摘要: 利用HPLC-MS/MS指导分离,从单花山竹子茎叶中分离得到4个新xanthones (oliganthins A-D)和1个已知的笼状xanthone (gaudichaudione H),它们的结构通过波谱方法得以鉴定。5个化合物均进行了诱导宫颈癌细胞HeLa-C3凋亡活性测试,抑制浓度在10 μ#25110;更低。另外,Oliganthins A,B和gaudichaudione H的诱导细胞凋亡活性再通过PARP(PARP是被激活的caspase-3的作用底物)的裂解反应由免疫印迹进行监测得到进一步确认。此外,通过MTT法还测定了五个化合物作用于HeLa-C3细胞的IC50值。其中gaudichaudione H的IC50值最低为0.90μ#65292;其他四个新化合物的IC50值分别是1.58,1.52,4.15,和7.82 μ#12290;结果表明 gaudichaudione H在这几个xanthone中有最强的诱导细胞凋亡和阻止细胞生长能力,具有开发成为一种新的抗癌先导化合物的潜能。

英文摘要: Four new compounds, oliganthins A-D (1-4), and one known caged xanthone gaudichaudione H (5) were isolated from the leaves of Garcinia oligantha guided by HPLC-MS/MS. All of the five compounds were evaluated for their apoptosis-inducing effects. All of them induced cell apoptosis at 10 μor lower concentrations. The apoptotic activity of oliganthins A, B and gaudichaudione H were further confirmed by detecting the cleavage of PARP using the method of Western blot. Moreover, the values of IC50 were measured for all four compounds on HeLa cells using the MTT assay.
