中文摘要: 本项目以一个植物特有的长链非编码RNA HID1为研究对象,基于前期对HID1分子参与红光信号介导的光形态建成的功能研究,进一步阐明HID1以RNA形式直接作为转录调控因子的作用机制。我们的初步研究结果提示,红光主要受体phyB和光形态建成正调控因子HY5可能与HID1共同参与了持续红光条件下光形态建成负调控因子PIF3基因的转录抑制,与激活PIF3基因转录的转录因子EIN3/EIL1作用相反。本项目拟改进并综合运用RNA纯化的染色质分离(ChIRP)和MS2标签化RNA成像等分子生物学技术方法,建立HID1与光形态建成关键蛋白因子协同或拮抗调控PIF3基因转录的分子关联。我们的目标是阐明HID1调控红光信号介导的光形态建成的分子机制,深化人们对于非编码RNA在光控植物生长发育中调控作用的认识,为阐明光形态建成中的转录调控网络提供新视角。

英文摘要: Taking HID1,a plant-specific long noncoding RNA,as our research focus, and based on our previous study on its functional analysis in red-light control of photomorphogenesis, we aim to study the molecular mechanism of HID1, a regulatory RNA, in transcriptional regulation. Our preliminary data showed that the major red light receptor phyB and photomorphogenic-promoting factor HY5 may work coordinately with HID1 in repressing PIF3 transcription, during which EIN3/EIL1 function in the opposite way. By using multiple molecular biology approaches including our modified Chromatin Isolation by RNA Purification (ChIRP) and MS2-tagged RNA imaging, we will figure out the cooperative and/or antagonistic relationships between HID1 and the key protein players in transcriptional control of PIF3 expression. The goal of this project is to elucidate the molecular mechanism of HID1 in the red-light control of photomorphogenesis. Our study will not only help to understand the regulatory role of ncRNAs in the light control of plant growth and development, but also provide new insights on transcriptional programming in photomorphogenesis.
