中文摘要: 肝癌是恶性肿瘤中常见且难以早期发现的一种,开展肝癌的早期诊断和治疗意义重大。我们拟利用激光诱导荧光全内反射显微术结合微流控芯片技术,开展肝癌细胞内基因和蛋白类肿瘤标志物的单细胞、单分子水平荧光检测方法研究,进行单个肝癌细胞内肿瘤相关抗原甲胎蛋白AFP mRNA、miRNA家族中mir-18的单分子荧光检测,单分子水平实时研究抗肿瘤药物诱导细胞凋亡过程中细胞膜表面肿瘤坏死因子受体时空变化。本项目所发展的单细胞内单分子检测方法具有检测灵敏度高、耗样量少、通量大的特点,我们期望基于此项研究能够在肝癌的早期诊断上取得进步,并进一步为肿瘤的预警和早期诊治提供新的有效方法与途径。

英文摘要: It makes great sense to carry out early diagnosis and treatment of liver cancer. In this study, we will develope an ultrasensitive single-molecule detection method for tumor markers (genes and proteins) in single HepG 2 cells using a combination of total internal reflection fluorescence microscopy (TIRFM) and micro-fluid chip. With this method, the Alpha-fetoprotein mRNA、mir-18 in microRNA families will be analysed quantitatively at single molecule level in single cells.Single-molecule detection of spatially variation of the tumor necrosis factor receptor (TNFR) at the living cell surface will also be carried out during the apoptosis induced by anti-cancer drug. The method developed in this research requires less sample, has high detection sensitivity and can implement high-throughput single-cell analysis. We hope this study can make improvement in the early diagnosis and treatment of liver cancer and provide effective method for the prewarning of tumor.
