中文摘要: 光学显微成像被广泛应用于生物医学、材料等领域,是现代科学不可缺少的工具。然而,传统的光学显微镜依赖于设计复杂的显微物镜,使得其视场范围较为有限,同时显微物镜的复杂设计也增加了系统成本。本项目计划对无物镜宽视场光学显微成像方法进行研究,探索在没有传统显微物镜的前提下重新设计光学显微系统,力求在保持类似分辨率的前提下得到比普通显微镜宽得多的视场,从而实现高通量显微成像。我们将研究利用聚焦点阵列对样品进行扫描成像的方法。通过对聚焦点阵列产生机理的理论模拟和实验研究,力求找到较优的制作聚焦点阵列产生器件的办法,以较好地实现具有与显微镜类似分辨率大小的大面积聚焦点阵列。然后我们将利用产生的聚焦点阵列进行显微成像实验,同时研究其中存在的扫描方法与图像重构等实际问题。上述研究将为类似的新型显微方法提供理论和实验基础,其中聚焦点阵列产生机理的研究也有可能被应用于其他相关的光学领域。

英文摘要: Optical microscopy has been widely used in biomedicine, material science, and many other areas. However, traditional optical microscope relies on complex specially-designed microscope objectives, which limits its field-of-view. Meanwhile, the complex design of the objective also increases the system cost. This project plans to study the methods for microscopy without using objectives, and performs research on re-designing the optical microscope system, in order to get a much wider field-of-view while maintaining similar resolutions, and thus achieve high-throughput microscopy. We will study the principles of using focal spots grid for imaging by scanning methods. Based on the theoretical and experimental study of mechanisms to generate the focal spots grid, we will try to find the optimal methods to fabricate the device for focus grid generation, so as to achieve a wide-area focus grid with similar spot size as the resolution of microscope. Then we will use the focus grid for microscopic imaging experiment, and study the practical issues related to scanning mechanisms and image reconstructions. The abovementioned research will provide theoretical and experimental guidance for novel microscopic methods.
