中文摘要: 研究小麦籽粒的发育过程,解析籽粒发育的调控机制,对于发掘小麦籽粒性状相关基因具有重要意义。miRNAs在植物生长、发育等各个过程中都具有十分重要的功能,目前国内外还未见系统研究miRNAs参与小麦籽粒发育过程的报道。前期工作我们已完成对小麦籽粒不同发育时期小RNA的高通量测序,并发现和鉴定了上百个在籽粒中表达的miRNAs。基于前期工作,本项目拟通过生物信息学分析和多种分子生物学手段,构建籽粒发育时期的miRNA表达谱,从中筛选并验证具有显著差异表达特征的miRNAs,并对其靶基因进行鉴定和分析。在此基础上,构建miRNAs参与小麦籽粒发育的整体作用模型,系统评价并找出调控籽粒发育的重要miRNAs,并通过转基因方法来证实代表性miRNAs及靶基因(包括miR156和TaGW8)在籽粒发育中的功能。预期的研究结果将完善小麦籽粒发育相关的调控网络,并有望为小麦分子育种提供新的基因元件。

英文摘要: It is crucial in discovering genes associated with wheat grain traits by studying the grain developmental process and resolving the regulatory mechanism of grain development. miRNAs play a key role in plant growth and development, however, there are rare reports on systematically studying miRNAs involved in the developmental process of wheat grains till now. In the preliminary work, we completed high throughput sequencing of small RNAs at different developmental stages of wheat grains, discovered and identified hundreds of miRNAs expressed in the grains. On this basis, this project aims to construct wheat miRNA expression profiles during the grain developmental process, and then select and identify differentially expressed miRNAs, confirm and analyze their target genes through bioinformatics analysis and molecular biology methods. According to these results, a miRNA working model linked with wheat grain development will be constructed, miRNA functions will be systematically evaluated, and important regulatory miRNAs involved in the grain development will be selected. For representative important miRNAs and their target genes (including miR156 and TaGW8), transgenic research will be performed to verify their functions in the grain development.
