中文摘要: 脂肪酸糖酯化合物是一类应用广泛、绿色环保的生物表面活性剂。本项目拟把溶剂设计和酶的选用及固定化两方面结合起来,建立一个高效、绿色的酶法合成糖酯的反应体系。一方面,依据离子液体设计的新理念来设计两类新型的绿色离子液体(铵盐/鏻盐离子液体与深度低共熔溶剂体系)作为反应介质;另一方面,开发国产的扩展青霉脂肪酶及其交联聚集的无载体固定化技术,并以此固定化酶作为催化剂。以脂肪酶催化糖酯合成为实例,研究分析离子液体的结构组成对反应物溶解度以及酶的催化活性、稳定性的影响及制约作用,探讨酶催化在深度低共熔溶剂体系中的应用及基本作用原理,为离子液体的理性设计提供理论和实验依据。此项研究对糖酯的绿色合成、新酶的应用开发、以及绿色离子液体的理性设计及其对生物催化的作用机制研究,无论在理论上还是在应用上都具有重要意义。

英文摘要: Sugar fatty acid esters are bio-surfactants with broad applications in food, cosmetic and pharmaceutical industries. The aim of this project is to establish a green reaction system for sugar ester synthesis with high efficiency. We are going to 1). design two types of advanced ionic liquids (ILs), ammonium and phosphorium ILs and deep eutectic solvents (DESs), as the reaction medium, based on the new guidelines we have proposed for IL design; and 2). develop the use of a new lipase, Penicillium expansum lipase, as a catalyst for sugar ester synthesis, and its immobilization based on the CLEA (cross-linked enzyme aggregates) technology. This research will provide valuable information both in theory and in applications for green synthesis of sugar esters, development of new enzymes, rational design of green ILs and investigation about their effect on biocatalysis.
