中文摘要: 麻疯树为大戟科多年生木本,重要的生物柴油能源作物和生态经济树种,分布种植于热带或亚热带南缘,属耐热喜温、低温敏感植物。低温冷害是麻疯树生产的主要灾害和种植区域的限制因子,麻疯树抗冷机制和耐寒品种的培育是生产实际中亟待研究和攻克的难题。我们利用酵母单杂文库技术,成功构建麻疯树低温响应转录因子基因文库,得到9大功能类群共184个基因的全长或片段cDNA。本项目从基因组学的高度揭示麻疯树低温应答转录因子家族内部及家族之间相关基因的作用模式及网络调控,发掘麻疯树抗寒耐冷性状形成的重要基因;采用常规基因克隆技术,分离耐寒转录因子(调控)基因;通过转基因拟南芥或麻疯树验证其功能和生物学作用,为麻疯树分子改良和抗冷新种质创制提供理论依据和技术保障。其目标是获得3-5个麻疯树重要耐寒转录因子基因和100株以上转抗冷基因麻疯树植株,申请国家发明专利1项,发表SCI学术论文2篇以上,培养研究生2名。

英文摘要: Jatropha curcas, a perennial oil plant belonging to the family Euphorbiacea, grows in the marginal and poor soil, it is widespread in tropical and subtropical areas. Nowadays, J. curcas is extremely valuable for hunman being due to high oil content in its seeds which can be reformed as biodiesel. However, it does not tolerant low temperature. Therefore, in this study, some transcription factor genes were isolated and deeply investigated in response to adverse environment signal, respectively. To gain a better understanding of changes in gene expression associated with cold acclimation in the woody oil plant Jatropha curcas, a Yeast one-hybrid library was constructed and screened. A full-length cDNAs library for use the yeast one hybrid system by SMATR technology was constructed from J.curcas seedlings under low temperature conditions. 184 clones with funcation annotation in 254 clones were obtained, in which, 15 transcription factors were related to cold tolerance.Isolated and cloned 10 cDNA of regulative and functional genes related to low temperature tolerance in J. curcas. Our studies have offered our knowledge on understanding the molecular mechanism of J.curcas responding to low temperature.
