中文摘要: 逆转录酶对病毒RNA的逆转录作用是癌症、艾滋病和白血病等重大疾病产生的重要毒理学基础。以抑制逆转录酶活性为机理的药物设计是当前抗肿瘤、抗艾滋病新药开发的重要组成部分。我们设计了一系列具有核酸碱基结构的单核和双核Salen金属配合物,一方面利用Salen金属配合物轴向配位能力与RNA结构中单链部位碱基中的电子给体形成配位键,另一方面依靠配合物结构中的碱基结构与RNA结构中的单链部位的碱基产生氢键作用。借助配位键和氢键的共同作用,使Salen金属配合物成为与当前研究中仅靠静电作用、疏水作用、氢键以及π#960;堆积等弱相互作用而与RNA或逆转录酶结合的逆转录酶抑制剂相比,具有更高亲和力的RNA结合试剂,从而更加有效的阻断逆转录酶对病毒RNA的逆转录作用。进一步设计了一系列结构各异的金属配合物,能够抑制DNA的转录过程,从而抑制肿瘤细胞生长。

英文摘要: Reverse transcription of virus RNA by reverse transcriptase is the essential toxicology basic of cancer, HIV and leukaemia. Design of new reverse transcriptase inhibitors is one of the most important parts of current antitumor and antiHIV drugs development. We have designed and synthesized a series of mono- and multi-nuclear Salen-metal complexes containing bases of nucleic acid structures. On the one hand, the complexes can bind RNA bases with axial coordinative function. On the other hand, the base-like structures in the complexes can bind RNA bases via hydrogen bonds. With the cooperation of these two binding modes, Salen-metal complexes may serve as RNA binding reagents with higher activities than binders currently used, which can only interact with RNA via only one mode, like electrostatic, hydrophobic, π#960; stack and hydrogen bonds, thus to inhibit the activity of virus RNA reverse transcription more effectively. A series of structurally diverse metal complexes have been further designed. They were found to be potent inhibitors towards the DNA transcription and effective antitumor reagents.
