中文摘要: 炎性体广泛存在于动物体内,能迅速触发免疫反应,在宿主的感染、免疫、疾病过程中都发挥着十分重要的作用。近年来,炎性体同疾病的关系成已为生命科学、医学研究的热点之一,而在兽医方面有关炎性体的研究不多,对炎性体通路与病毒感染、免疫调节及动物疫病的关系及作用所知甚少。鉴于此,本项目根据炎性体的最新研究成果,结合猪瘟病毒的感染致病特点,针对猪瘟病毒感染致病、免疫逃逸机制尚不清楚等科学问题,在我们前期研究的基础上,以猪瘟病毒/编码蛋白与细胞炎性体信号途径相互作用为主线,从猪瘟病毒体内/外感染入手,选择相关细胞、炎性体分子、凋亡及细胞因子等作为研究对象,结合炎性体通路调节剂、基因功能干预等手段,从不同层次和角度进行研究,以揭示炎性体对猪瘟病毒感染、免疫调控的分子作用机制,使我们更加明确猪瘟病毒感染对机体的损伤新机制, 进而从新的角度了解猪瘟病毒的感染致病机制,为猪瘟防控寻找新途径、提供新策略。

英文摘要: Inflammasome, as a key component of cytosolic surveillance of the innate immune system, plays a important role in host infection, inflammation,occurrence and development of various diseases, as well as the immunological process. In recent years, the relationship between inflammasome and the diseases has become a hot spot in the life science and medical research. However, few studies related to inflammasome have been done in veterinary and animal husbandry. Furthermore, correlation and roles of inflammasome in animal diseases, viral infection and immunological regulation are prooly understood. In view of these, based on our previous studies and according to the theory of inflammasome, the lastest findings and the features of classical swine fever virus (CSFV) development, the project aims to reveal molecular mechanisms of pathogenicity and immune regulation of inflammasome in CSFV infection. After CSFV infection in vivo and vitro, the relevant cells, molecular marker of inflammasome, apoptotic factors and cytokines are investigated using agonists/inhibitors or gene interference. In this study, the molecular mechanisms of inflammasome in immune regulation after CSFV infection are fully investigated from different levels and angles. It clearly indicates a new mechanism of the damage of CSFV infection to the host body, making us to understand the pathogenesis of CSFV in a new perspective, bringing a new way to enhance the immune effects of CSFV, and providing new strategies and new ideas for prevention and control of classical swine fever.
