中文摘要: H9N2禽流感病毒在多种陆禽流行并反复感染哺乳动物猪和人,其对公共卫生安全呈潜在巨大威胁,故探讨H9N2病毒如何跨种间屏障传播人,筛选病毒跨种间屏障传播的特异分子标记具有重要意义。我们前期研究发现,陆禽呼吸道均表达禽流感病毒 SAα-2,3 Gal和人流感病毒SAα-2,6Gal受体,提示陆禽在流感病毒生态系统中可能也是重要中间宿主, 有助于病毒基因进化,形成感染人的潜能。本项目在此研究基础上,首先用鸭 H9N2流感病毒在陆禽(鹧鸪、火鸡)反复复制后形成的变异毒株,然后将该变异毒株体内感染猪、体外感染离体人呼吸道组织,观察其在猪各组织器官和人呼吸道感染情况,同时应用基因测序法,比较变异病毒H9N2与亲代鸭病毒H9N2各基因片段的变化,筛选 H9N2 病毒潜在感染哺乳动物的基因变化特别是特异分子标记。此研究结果为探索陆禽在流感病毒跨种间传递中的作用、疫苗的研制和流感病毒的监测提供分子学基础。

英文摘要: H9N2 avian in?uenza viruses have been circulating in multiple terrestril birds and repeatedly infecting mammals,including swines and humans, posing a signi?cant threat to public health potential. Therefore, it is important significance to explore H9N2 avian influenza virus how to cross interspecies barriers from avians to humans and screen these specific molecular markers.Our previous study showed that both SAα2,6Gal and SAα2,3Gal were expressed in terrestrial birds. These findings suggest that terrestrial birds themselves may also act as important intermediate host in ecosystems of influenza viruses to facilitate gene evolution and generate an influenza virus with infecting humans potential.On the basis of previous study, we develop a variant virus H9N2 through repeated replication in terrestrial birds,including chukar and turkey, then perform variant virus H9N2 in vivo infection of swines and ex vivo infection of surgically removed respiratory tract tissues of human and observe infection of variant virus H9N2 in all tissues and organs of swine and respiratory tract tissues of humans.
