中文摘要: 几丁质合成是昆虫的一个重要生理过程。目前,昆虫几丁质合成的调控途径主要包括:利用RNAi技术研究几丁质合成通路基因、激素以及microRNA对几丁质合成的调控等。除了以上已有报道的调控因子,目前尚未有任何实验结果显示所有参与昆虫几丁质合成的正、负调控元件及相关影响网络。本项目以水稻害虫褐飞虱结合果蝇细胞为研究对象,首先利用果蝇全基因组dsRNA的高通量筛选获得影响几丁质合成的所有调控元件,然后利用水稻害虫褐飞虱验证上述初筛结果,并在昆虫活体中鉴定重要元件在昆虫生长发育和几丁质合成中的功能,最后初步建立调控昆虫几丁质合成的交互网络。该项目不仅可在理论上阐明几丁质合成中的重要调控元件及其功能,而且可为利用RNAi来控制害虫奠定基础。

英文摘要: Metamorphosis is an important developmental process in insects. At present, the chitin biosynthesis was regulated by RNAi of the chitin biosynthesis pathway genes, hormone receptor and microRNAs. Besides of the regulated factors that reported aboved, none of results about positive and negative factors and related networks that involved in the chitin biosynthesis have showed yet. Focusing on Nilaparvata lugens with Drosophila melanogaster cells as the research objects, three aspects of researches were conducted. First, all of regulated factors affected the chitin biosynthesis will be acquired through genome-wide RNAi screen. Second, the key hits will be verified in Nilaparvata lugens through living injection to identify the function of growth and development. The last, the networks along with the effect factors that regulated the chitin biosynthesis will be establish preliminarily. This project will clarify the key factors in chitin biosynthesis in theory, and also be an basic on pest control through RNAi.
