中文摘要: 以不同种属纤毛虫为研究对象,以本研究室自创的扫描电镜样品制备方法为核心技术,去除纤毛虫体内细胞质,使其表膜外翻,皮层下结构完整暴露,从而更加清晰展示纤毛虫皮层纤维系统的三维结构和特征;同时结合免疫荧光实验,观察中间纤维蛋白抗原的表达,以确定类中间纤维的数量和空间分布;辅以一般形态学和电镜观察;最终,通过比较和分析不同种类纤毛虫皮层纤维系统超微结构和免疫荧光实验结果,达到系统全面地揭示纤毛虫皮层纤维系统的特征、性质和组成的目的,从而探讨纤毛虫类中间纤维的分布及其生物学功能,为高等动物中间纤维研究提供新的参考与借鉴。

英文摘要: To take different species of ciliates as research subjects, a made-up scanning electron microscopy sample preparation method for the core technology,to remove the cytoplasm of these ciliates, reverse their membranes and completly expose their subcortical structures. Then to show clearly the three-dimensional structure and characteristics of the ciliates cortical fiber system. Additional to observe the intermediate filament(IF) protein antigen expression by immunofluorescence experiments to determine the quantity and the spatial distribution of the intermediate-type filament. Supplemented by normal morphology and electronic microscopy observation. Finally, achieve to reveal the nature, characteristics and composition of ciliates cortex fiber system through comparison and analysis of different kinds of ciliate cortical fiber system ultrastructure and immunofluorescence results, to explore ciliates intermediate-type fiber distribution and biological functions, to provide new informations and references for the research on intermediate filament of higher animals.
